You need to make it clear your busy. There are lots of subtle ways to do this without offending your colleagues. For example when they come in and start chatting pretend you were just on your way out the office. The people who have nothing better to do during the day than chat for 45 minutes will often move on to someone else to bother. You can then walk back to your desk within 2-3 minutes and continue your productive day.

Oh dear. This is a tricky one. I think you just need to do your best to avoid this person. If they are not your boss go to your boss and let them know they are taking up your time during the day. Let your boss know you feel you would get more done if this person didn't distract you everyday.
- your colleague who actually who interfered in your personal issue and screwed you up, but still you have to work with him.
Try not to be that person within your company who complains about others. Don't worry to much about this person. Focus on your own job and do your lob amazing every time. You will get noticed by others within your company and hopefully promoted way beyond the irritating person your dealing with right now.
